Noi structuri peptidice cu potențiale aplicații în terapia țintită și diagnosticul timpuriu în cancer

New peptides for targeted cancer therapy and early diagnosis

Obiectivul general: Obiectivul general al proiectului constă într-o strategie in doi pași, primul reprezintă proiectarea unor structuri peptidice noi cu un grad ridicat de permeabilitate pentru membranele celulare și specificitate pentru receptorii celulelor canceroase și al doilea, folosirea acestor noi molecule pentru a ținti proteine ale căilor mecanosenzitive.

The main objective of this project proposal is built around a two steps strategy, first to design peptide structures with high degree of cell membrane permeability and specificity for protein receptors overexpressed in cancer cells and secondly, to target proteins from mechanosensitive pathways of cell migration with these new molecules.

Rezumat: Deși în ultimii ani s-au făcut progrese remarcabile, cancerul încă reprezintă o provocare majoră pentru comunitatea științifică. Mai ales, detectarea celulelor canceroase în stadii incipiente de dezvoltare este încă problematică. Descoperirea unor noi peptide cu capacitate mare de penetrare a membranei plasmatice și specificitate pentru receptorii supraexprimați în tumori, prezintă un interes deosebit atât pentru detectarea cât și pentru dezvoltarea de noi sisteme de administrare țintită a medicamentelor. Peptidele care au specificitate pentru celulele tumorale dar și proprietăți de penetrare a membranelor celulare reprezită cele mai eficiente sisteme pentru eliberarea țintită: dimensiuni mici, costuri reduse de producție, flexibilitate în posibilitățile de conjugare. Din nefericire, în cazul celor mai multor terapii, medicamentele sunt introduse în celulă prin endocitoză și apoi sunt blocate în endosomi, inainte de exercitarea efectului terapeutic.

Scopul acestui proiect este de a combina specificitatea peptidelor de tip homing cu peptide care au proprietatea de a penetra membranele celulare, pentru a genera noi instrumente moleculare cu un grad ridicat de specificitate pentru celule canceroase și permeabilitate a membranei celulare. Metode precum microscopia de fluorescența, electrofiziologie, spectroscopie, tehnici de biologie moleculară vor contribui la înțelegerea mecanismului de internalizare și interactiune cu proteine mecanosensitive al noilor peptide.

Abstract: In spite of the remarkable progress in the past years, cancer still represents a major challenge for scientific community. Especially, cancer cell detection in early stages of development is still problematic. Discovery of new peptides with high penetration capability of plasma membrane and specificity for receptors overexpressed in tumors is of particular interest for both, detection and development of new drug delivery systems. Tumor-homing peptides have a few properties over other systems that make them the best candidates for specific targeting: small size, low cost of production, flexibility in conjugation possibilities. Unfortunately, for most targeted therapies, the macromolecular drugs are taken into the cell through endocytosis and then end-up trapped into endosomes.

The goal of this project is to combine the specificity of homing peptides with special properties of membrane permeability of cell penetrating peptides and antimicrobial peptides, respectively, to generate new molecular tools with high degree of specificity and cell membrane permeability. Methods like fluorescence and super-resolution microscopy, electrophysiology, spectroscopy, cellular and molecular biology techniques will shed light into understanding the uptake mechanism, membrane trafficking and interactions with proteins of mechanosensitive pathways of cell migration of the new peptides.

Obiectivele proiectului:

Obiectiv 1: Selecția peptidelor homing, penetrante și antimicrobiene, proiectarea noilor structuri peptidice cu proprietăți penetrante și de selecție îmbunătățite

  • O1.A1. Selecția peptidelor homing cu specificitate pentru receptori ai celulelor canceroase și construcția noilor molecule peptidice cu structuri penetrante sau antimicrobiene.
  • O1.A2. Sinteza și marcarea fluorescentă a noilor peptide cu fluorofori adecvați

Obiectiv 2: Studiul proprietățior penetrante și verificarea specificității noilor peptide pentru recepri ai celulelor canceroase.

  • O2.A1. Prepararea membranelor model care mimează membrane celulelor normale, canceroase și a endozomilor.
  • O2.A2. Investigarea acumularii și/sau internalizării în membranele model a peptidelor marcate fluorescent.
  • O2.A3. Influența pH-ului asupra internalizării și procesului de formare de pori.
  • O2.A4. Rolul moleculelor tip „cargo” in translocarea peptidelor.
  • O2.A5. Reconstituirea receptorilor celulelor canceroase în proteoliposomi și fuzionarea cu membranele biomimetice.
  • O2.A6. Determinarea selectivității noilor peptide pentru receptorii celulelor canceroase

Obiectiv 3: Investigarea interacțiunilor moleculare ale peptidelor selectate cu proteine incipiente ale căilor mecanosenzitive ale migrației celulare/aderențe focale incipiente (nascente).

  • O3.A1. Expresia și purificarea proteinelor recombinante utilizate in cadrul proiectului.
  • O3.A2. Investigarea efectului peptidelor asupra cineticii de polimerizare și/sau depolimerizare a actinei.
  • O3.A3. Investigarea rolului peptidelor asupra activității proteinelor din căile mecanosenzitive ale migrației celulare

Obiectiv 4: Coroborarea rezultatelor obținute pentru activitatea peptidelor în vederea optimizării structurilor peptidice sau proiectării unor noi molecule eficiente, cu proprietăți penetrante îmbunătățite și specificitate ridicată pentru receptorii celulelor canceroase

The specific objectives and activities:

Objective 1: Selection of homing peptides, cell penetrating and antimicrobial peptides, design and labeling of the new peptides with improved penetrating and targeting properties.

  • O1.A1. Selection of homing peptides based on cancer receptor specificity and construction of new peptidic molecules with cell penetrating or antimicrobial peptide structures.
  • O1.A2. Synthesis and labeling of the new peptides with the appropriate fluorescent dyes.

Objective 2: Assessment of cell membrane permeability and specificity of the new peptides for cancer cell receptors

  • O2.A1. Preparation of model membranes mimicking the normal cells, cancer cells and endosomes
  • O2.A2. Investigation of accumulation and/or internalization in model membranes of fluorescently labeled peptides.
  • O2.A3. pH influence on peptide internalization and pore formation.
  • O2.A4. Role of cargo in peptide translocation.
  • O2.A5. Reconstitution of cancer receptors in proteoliposomes and fusion with model membranes.
  • O2.A6. Screening the selectivity of new peptides candidates for cancer cell receptors.

Objective 3: Investigation of molecular interactions of selected peptides with proteins of primary proteins of mechanosensitive pathways of cell migration/ nascent focal adhesions.

  • O3.A1. Expression and purification of the recombinant proteins used in this proposal.
  • O3.A2. Investigation of the effect of the peptides on the kinetics of actin polymerization and/or depolymerization.
  • O3.A3. Investigation of the effect of the peptides on the activity of proteins of mechanosensitive pathways of cell migration.

Objective 4: O4.A1. Corroboration of all gathered results on the activities of peptides in order to optimize the existing peptides or to design new efficient molecules with improved penetrating properties and high specificity for cancer cell receptors.
